Relation between Toll Like Receptor-2 Polymorphism and Helicobacter pylori Associated Peptic Ulcer Disease

Abstract
Background/Aims: Helicobacter pylori (H. pylori) infection is one of the most important causes of peptic ulcer disease. Recently, Toll-like receptor (TLR) 2, but not TLR4 is required for H. pylori lipopolysaccharide induced nuclear factor-kappa B activation. Therefore, it is important to investigate the relation between TLR2 polymorphism and peptic ulcer disease for the understanding of peptic ulcer disease pathophysiology. This study aimed to investigate the relation between the TLR2 polymorphism and the incidence of peptic ulcer disease in H. pylori infected Korean patients.

Methods: Blood samples were collected from a total of 190 H. pylori infected patients (gastric ulcer; n=74, duodenal ulcer; n=68 and control; n=48). DNA was extracted from buffy coat by using commercially available extraction kit. TLR2 Arg753Gln polymorphism was genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).
Results: Although we found TLR2 PCR product in all cases, TLR2 Arg753Gln polymorphism was not detected in this study subjects.

Conclusions: In conclusion, the polymorphism of TLR2 (TLR2 Arg753Gln) does not seem to be associated with pathogenesis of H. pylori related peptic ulcer disease in Korean patients. (The Korean Journal of Helicobacter and Upper Gastrointestinal Research 2008;8:20-25)
